Anti-Aging Interventions: Clinical Evidence & Research
According to Wikipedia, life extension represents concept of extending human lifespan through scientific methods, including dietary interventions, pharmaceutical compounds, and lifestyle modifications. Human aging constitutes complex, multidimensional process characterized by gradual, progressive loss of physiological integrity and cellular functions.
Aging constitutes recognized major risk factor increasing susceptibility to diseases, especially in elderly adults. Functional and biological decline remarkably affects body systems. Interest in therapeutic potential of nutritional supplementation for stimulating longevity and reducing risk of aging-related pathologies has become prominent focus in clinical research.
Eight Promising Anti-Aging Compounds
Comprehensive review (PMID: 38181790) summarized current knowledge on eight promising drugs and natural compounds tested clinically: metformin, NAD+ precursors, glucagon-like peptide-1 receptor agonists, TORC1 inhibitors, spermidine, senolytics, probiotics, and anti-inflammatories. Multiple clinical trials commenced evaluating efficacy against age-associated diseases including diabetes, cardiovascular disease, cancer, and neurodegenerative diseases.
Reasonable expectations exist that drugs able to decelerate or reverse aging processes will exert broad disease-preventing or -attenuating effects. Outcome of past, ongoing, and future disease-specific trials may pave way to development of new anti-aging medicines. Drugs approved for specific disease indications may subsequently be repurposed for treatment of organism-wide aging consequences.

Natural Methods โDietary Supplements and Natural Products
Systematic review (PMID: 37441528) examined natural products from food sources classified as antiaging compounds. Polyphenols, saponins, alkaloids, and polysaccharides promote health and prolong life via various mechanisms. Sirtuin, AMP-activated protein kinase, mammalian target of rapamycin, p53, and insulin/insulin-like growth factor-1 signaling pathways are most widely studied.
These compounds delay aging development by mitigating cell proliferation, DNA damage, oxidative stress, inflammation, and activating cell autophagy and mitochondrial homeostasis. Additional translational evidence with robust study design on tolerability, safety, effectiveness, and off-target effects in aging population would enhance clinical potential.
Resveratrol Clinical Evidence
Clinical trial (PMID: 35950117) investigated resveratrol effects on skin health parameters. Research demonstrated resveratrol accelerates wound regeneration and healing processes, diminishes scarring, and reduces scar size. This occurs through increased concentration of endothelial growth factors, consequently leading to narrowing of wound edges. Resveratrol protects skin against harmful effects of type B ultraviolet radiation, which represents main factor in skin aging processes.
Systematic review of resveratrol clinical trials identified almost 200 studies evaluating resveratrol across at least 24 indications over past 20 years, including cancer, menopause symptoms, diabetes, metabolic syndrome, and cardiovascular disease. However, currently no conclusive clinical evidence exists to advocate recommendation in any healthcare setting.

Implementation Challenges and Future Directions
Review of longevity-promoting supplements (PMID: 37278035) noted resveratrol shown to decelerate aging and cellular senescence in animal models. However, no successful clinical trials yet completed in humans regarding dose limitations. Potent anti-aging and anti-senescence efficacy documented in several in vivo animal models. Chronic resveratrol treatment might result in nephrotoxicity, requiring attention to safe dosage and duration.
Substances with potent anti-aging properties not historically considered by FDA and regulatory agencies as suitable candidate drugs for clinical trials, because agencies did not recognize aging as clinical condition. However, increasing recognition that aging generally accompanies health issues associated with clinical conditions including sarcopenia, osteoporosis, atherosclerosis, hypertension, and dementia.
Safety & Complementary Approaches
Age-related energy decline requires comprehensive evaluation beyond supplementation alone. NAD+ precursor research demonstrates promising results in specific populations requiring further clinical validation.
Mitochondrial function and cellular energy optimization represent active research areas with emerging clinical evidence. Gender-specific interventions address unique physiological changes requiring tailored approaches.
